I have removal sound turned on in s pen settings but still no sound when I remove the s pen?

Please check volume settings: Settings -> Sounds and vibrations -> Volume

and slide the System and Notifications volumes accordingly.

Try again and see if you get sound when removing the S-pen.

Make sure do not disturb isn't on. Could try a restart on your device.

Check the settings relevant to your spen too please. 

Goodlock and it's plug in called pentastic also provides some amazing sounds.

Yes I have ensured do not disturb is turned off, all relevant settings are turned on to allow sound, also setup on goodlock, restarted phone and still doesn't work

Reset your sPen in the settings first and if no joy then try the below option. 

Clear Cache Files from the phone's System Partition Section  > https://www.samsung.com/uk/support/mobile-devices/how-do-i-clear-the-cache-on-my-device/ You may need to insert your earphones or hook upto a powered On Laptop or use a Tv's USB Port to access the Clear Partition Section. This may help with lag or stutters as this clears the phone of old/unused or broken cache files.There is also a Repair Apps option in the Menu System too.

And just to add I assume your volumes are turned up and no case is hindering the speakers.
