I've had such issues and it seem to be very random. Sometimes tilting the phone forward cuts off the ear piece speaker and sound would come only from the bottom speaker. Tilting the phone back and forth would cause the sound to shift but not always.

Speakerphone calls

To prevent auditory shock and hearing damage, the upper speaker is disabled for speakerphone calls if the Proximity sensor is blocked. If you are not holding the phone to your ear, remove anything that may be blocking the sensors at the top of the phone.
