An update to previous response as Amazon prime is now available directly on SkyQ, HDR10+ or Dolby Vision (on applicable sets) will not be supported on this platform though unlike the Tv apps. https://www.forbes.com/sites/johnarcher/2020/12/14/sky-q-and-now-tv-get-amazon-prime-video-support

It just plugs into a hdmi port on the tv. Doesn’t matter really which one.

Hi @dereksimpson yes as @Daveyboy123 has said any HDMI port should be fine.
Process pretty straightfoward but shall include a quick guide here. https://www.hellotech.com/guide/for/how-to-set-up-amazon-fire-stick

Hi @dereksimpson Customers will need to change the input to access the services on the Firestick connection. It will not show on the Sky connection. For instance if the Sky Box is connected to HDMI1 users would need to select the appopriate input for the Firestick whether it be 2 ,3 or 4 depending on your TV.
on the other hand if you are specifically referring to Amazon Prime it is available as an app on Samsung Smart Tvs, it is not integrated into the Sky service in the same way Netflix is, though only available via SkyQ. Sky though has no deal with Prime on any of its platforms.
